  Effect of closed fracture on spot urinary thiobarbituric acid reactive substance level in old rats

  BACKGROUND: It has been unclear whether spot urinary TBARS level is increased in the healing process of closed fractures in vivo. Therefore, the present study aimed to clarify the effect of closed fracture on spot urinary thiobarbituric acid reactive substance (TBARS) level in old rats.
METHODS: Eight male Wistar rats aged approximately 1 year were used in this study. Four rats were artificially fractured at the left fifth metatarsal bone (Fr rats). Other rats were received sham maneuver for fracture (sham Fr rats). Spot urine samples from all of the rats were taken in separate cages before and after fracture or sham maneuver daily for 1 week. TBARS and creatinine (CR) concentrations in spot urine samples taken on each day were analyzed.
RESULTS: In the cross-sectional analyses, the TBARS nmol/CR mg ratio at 3 and 4 days after fracture was significantly higher in the Fr rats (p < 0.05) than in the sham Fr rats at the same time points. Moreover, in the longitudinal analyses, TBARS nmol/CR mg in the Fr rats significantly increased at 3 and 4 days from the fracture (p < 0.05) compared with before fracture.
CONCLUSIONS: These results suggest that increased oxidative stress in vivo was induced by closed fracture, which is reflected in spot urine specimens. Therefore, to analyze spot urine samples after closed fracture in rats may be useful in investigations on therapeutic strategies for fracture.
